Effect of pregnancy-specific β1-glycoprotein on myeloid-derived suppressor cell differentiation

Myeloid-derived suppressor cells (MDSCs) are a heterogeneous cell population that primarily suppress T lymphocytes in healthy pregnancies and pathologies. MDSCs one of the key regulators immune responses. Finding ways to control them is important for treatment cancer, autoimmune diseases, miscarriage, post-transplant complications. The mechanisms suppression by MDSC are: expression CD73, ADAM17, PD -L1, production Arg 1, iNOS, IDO, IL -10 TGF-b1. Pregnancy-specific b1-glycoprotein (PSG) has modulatory effects on dendritic macrophages mediate shift phenotypes toward Th2 Treg. We have previously shown native PSG suppresses Th17 differentiation cytokine production, stimulates IDO monocytes Tregs. Considering immunomodulatory properties role pathologies, aim our work was investigate effect recombinant vitro . were differentiated from CD11b + peripheral blood cells. CpG oligonucleotides stimulate via TLR9 and enhance anti-tumor immunity, an effect attributed to the activation of NK and CD8(+) T cells. Our recent work demonstrates that CpG ODN also induce monocytic myeloid-derived suppressor cells to mature into M1 macrophages, further aiding tumor elimination. This provides insight into the mechanism through which CpG promote tumor regression.
